چکیده

Abstract Study question Do the main psychoactive phytocannabinoid delta-9-tetrahydrocannabinol (THC) and its non-psychoactive analog cannabidiol (CBD) affect human sperm function? Summary answer THC reduces ligand-dependent activation of sperm-specific Ca2+ channel CatSper affects functions in vitro. What is known already Marijuana (Cannabis sativa) one most commonly used recreational drugs worldwide. Although impact phytocannabinoids on reproductive health has been investigated, there no evidence a direct effect function. design, size, duration We studied effects phytocannabinoid, THC, analog, CBD, as well their major metabolites influx via spermatozoa. CBD were selected to further evaluate action progesterone-, prostaglandin-, pH-induced CatSper. The hyperactivation, progressive motility viscous media, acrosomal exocytosis also assessed. Motility assessment performed Computer-Assisted Sperm Analysis (CASA). penetration into media assessed modified Kremer test. Acrosomal evaluated by flow cytometry Pisum sativum agglutinin-stained Main results role chance Both suppress natural ligand-induced calcium In particular, inhibits progesterone-induced at pharmacologically relevant concentrations non-competitive manner addition, impaired hyperactivation induced spontaneous Limitations, reasons for caution This an vitro study. The CatSper channel: a polymodal chemosensor in human sperm.

The sperm-specific CatSper channel controls the intracellular Ca(2+) concentration ([Ca(2+)](i)) and, thereby, the swimming behaviour of sperm. In humans, CatSper is directly activated by progesterone and prostaglandins-female factors that stimulate Ca(2+) influx. Other factors including neurotransmitters, chemokines, and odorants also affect sperm function by changing [Ca(2+)](i). Several liga...

Inhibition of the CatSper Channel and NOX5 Enzyme Activity Affects the Functions of the Progesterone-Stimulated Human Sperm

Background: Low levels of reactive oxygen species (ROS) and calcium are necessary for sperm function. NADPH oxidase 5 (NOX5) is a membrane enzyme which produces ROS. This enzyme is dependent on calcium for its activity. We investigated the importance of NOX5 and an important calcium channel (CatSper) on sperm function. Methods: This laboratory in-vitro study was done in Shiraz, Iran, 2016. Norma...

Regulation of the sperm calcium channel CatSper by endogenous steroids and plant triterpenoids.

The calcium channel of sperm (CatSper) is essential for sperm hyperactivated motility and fertility. The steroid hormone progesterone activates CatSper of human sperm via binding to the serine hydrolase ABHD2. However, steroid specificity of ABHD2 has not been evaluated. Here, we explored whether steroid hormones to which human spermatozoa are exposed in the male and female genital tract influe...

The CatSper calcium channel in human sperm: relation with motility and involvement in progesterone-induced acrosome reaction.

STUDY QUESTION Does CatSper have a role in the achievement of human sperm motility and in the Progesterone (P)-induced acrosome reaction (AR)? SUMMARY ANSWER CatSper1 expression is associated with human sperm progressive motility and the P-induced AR; it may have a role in the pathogenesis of asthenozoospermia. WHAT IS KNOWN ALREADY Knockout mice for any of the Catsper family genes fail to ...

The CatSper channel controls chemosensation in sea urchin sperm.

Sperm guidance is controlled by chemical and physical cues. In many species, Ca(2+) bursts in the flagellum govern navigation to the egg. In Arbacia punctulata, a model system of sperm chemotaxis, a cGMP signaling pathway controls these Ca(2+) bursts. The underlying Ca(2+) channel and its mechanisms of activation are unknown. Here, we identify CatSper Ca(2+) channels in the flagellum of A. punc...
